Salvesterols are undoubtedly one of the most important anti-cancer and anti-aging compounds yet discovered. As an anti-cancer agent, Salvesterols help to block many reactions associated with cancer.  Salvestrols are unique because it battles cancer in all three phases of the cancer process: initiation, promotion, and progression. When a person with a healthy immune system develops cancer cells, the body will deal with and dispose of mutant cells on a regular basis.  Cancer develops in the out-of-balance organism. Cancer cells create a protective shell around themselves (the cytochrome P450 enzyme, CYP1B1) which prevents the immune system from destroying them. 

Pancreatin is an enzyme normally secreted by the pancreas. The level of this enzyme has been found to be severely deficient in cancer patients. The pancreatic enzyme is one of the body's natural defenses against cancer.  Pancreatin serves to strip the protective enzyme from the mutant cancer cells, allowing the immune system to attack and destroy the cell.  Salvesterol is similar but has a secondary benefit as it is able to penetrate the barrier, then metamorphoses to become a deadly poison against the same cancer cell it has come in contact with.

The Salvesterols in Natural Edge's Salvestrin act as a protease inhibitor and slow down the late stages of cancer progression in a way similar to chemotherapy, but in a non-toxic fashion. Moreover as an antioxidant it counteracts the super-charged free radicals that ordinarily damage normal cells.  Salvestrin as a protease inhibitor blocks or reverses DNA damage within normal cells to stop them from mutating into a malignancy.

In March of 2002, the research of Professor Gerry Potter, demonstrated the cancer inhibiting benefits that Salvesterols can provide. Salvesterols when in contact with the enzyme CYP1B1 (which is only found in cancer tumors) cause a metabolic reaction to occur, and the powerful antileukaemic agent piceatannol is produced. Piceatannol is a cancer cell destroyer. Salvesterols are only active against cancer cells; they are very selective and non-toxic to healthy cells.

Professor Potter has described Salvesterols as ‘a wolf in sheep’s clothing’. It is likely that we have discovered a mechanism that can tackle cancer cells whether they are formed or forming. Salvesterols because they require enzymatic activation are known or described as a pro-drug, with the ability to avoid the serious side-effects of most commonly used, anti-cancer drugs that often severely damage normal tissue as well as in some cases rendering the user infertile.

Capitalizing on the CYP1B1 Enzyme

The cytochrome P450 enzyme, CYP1B1, is common to a wide array of cancer cells (leukemia, lymphoma, tumors of the brain, breast, prostate, lung, colon, stomach, kidney, etc.) and is not present in healthy tissue. The recent discovery of a new class of molecule (Salvesterols), and its important activity in removing diseased cells from the human body, are being reviewed. Salvesterols are non-toxic until activated by the CYP1B1 enzyme to induce apoptosis (death) in diseased cells. Tumor selectivity and the bioavailability of Salvesterols, and the implications of their discovery for treatment of disease, have been examined in detail.

Magnolol induces apoptosis in human leukemia cells via cytochrome c release and caspase activation.

Magnolol, was found to prevent proliferation of human HL-60 cells and Jurkat T leukemia cells via inducing cancer cell death. By contrast, Magnolol did not cause cell death in normal healthy cells. Polymerase was found during cell deaths induced by magnolol. In addition, both pan-caspase and selective caspase-9 inhibitor blocked magnolol-induced cell death. The cell death could also be partially attenuated by caspase-3 and -2 inhibitors. Magnolol induced the reduction of mitochondrial transmembrane potential and the release of cytochrome c into cytoplasm. In conclusion, our findings indicate that magnolol-induced apoptotic signaling is carried out through mitochondria alternations to caspase-9 and that then the downstream effector caspases are activated sequentially.
